Sequence motifs and prediction model of GBE editing outcomes based on target library analysis and machine learning

Base editor techniques were developed for precise base conver-sion without requiring double-stranded DNA breaks(DSBs)or an editing template(Komor et al.,2016;Nishida et al.,2016).Currently,base editors that can perform cytosine(C)to thymine(T)(cytosine base editors,CBEs),adenine(A)to guanine(G)(adenine base edi-tors,ABEs)(Gaudelli et al.,2017),and cytosine(C)to guanine(G)base editing(glycosylase base editors,GBEs)(Chen et al.,2021;Kurt et al.,2021;Zhao et al.,2021)are available for mammalian cells.The nucleotide sequence around the editing position,in or near the target area,is considered the key factor that determines the effi-ciency of base editing.
